I don't think linda hunt qualifies, since this is not a case of a male character being changed to female but instead it is an actress playing a male character, since, according to peter weir, he simply could not find a male actor who fit the bill. the gender of the character remained unaltered.
He Who Remains / Kang / Immortus - cast as Jonathan Majors, white in the comics. Currently appeared in the Loki MCU TV series, and already cast for Ant-man, and possible other properties.
